When I try to type the shortcut keys I want to use (alt + s for the section
symbol), nothing happens and the "assign" button is blanked out. The
dialogue box will only accept certain function keys like "insert" and
"delete, " the F2 and F3 keys. I should be able to type these keyboard
entries in, shouldn't I?

Where are you doing this?

If you open Insert Symbol and select the section sign character, it
lets you type "Alt-s" as the keyboard shortcut.
