You can't search the Time Machine backup drive directly using Spotlight (or Finder's Find command, or other software which uses the Spotlight index). You can search the backup while in the "outer space" Time Machine view. My preferred method: Start by using Cmd-F in Finder, enter the search criteria you want. The location to search should be either "This Mac" or the folder on your MAIN hard drive where the item was originally located (or a parent folder, if you don't remember exactly where the file was). Then use the "Enter Time Machine" command (e.g. from the Time Machine status menu, or the Dock icon if you haven't got rid of it). You can then use the up-arrow button in the "outer space" view to step backwards in time, or the timeline on the right if you remember roughly when the file existed. If you use the up-arrow and down-arrow buttons, Time Machine usually jumps backwards until the search results change, but I've found it sometimes needs some extra nudging.
