I have a user that is loading video files in a folder under their desktop
(ie. desktop\video files). Does windows load everything in the desktop
folder and subfolders into memory on bootup? Or is it just the "desktop"
folder itself that needs to stay "lean".

You'd be better posting in an appropriate newsgroup - Windows XP
newsgroup? (you don't even mention the OS in question).
