I am prepping a Dell T610 and want to add in an eSATA card so i can
use 1 TB eSATA drives as backup...does anyone have experience using
eSATA w/SBS or the T610? I don't really know what card would work
best or what i'm in for in terms of getting it working, at least so it
is Hot Swapable...

It will work just fine.

An eSata drive behaves much like any internal drive. In fact, I have
cables to hook up internal drives to eSata interfaces and vice versa.

I have esata on SBS03 servers, if the drives are connected when the
server reboots, the server will hang with OS not found - I believe this
is a BIOS issue, since the drives are changed daily, if they are not the
same drive as when the server was booted they see them as new drives and
try to boot from them.

I just installed a T610 with SBS and 1.5TB of disk space, using "USB"
external Seagate 1.5GB drives, takes about 1.5 hours for first backup
then 20 minutes for each backup after that - we do 6 backups per day.

You don't need eSata performance in SBS 08 like you did in 03.

So, you have an eSATA working in Hot Swap mode so you can remove a
drive and swap it with a new one each day? That is my biggest concern
- Leonid mentioned it is seen as an internal drive, but an internal
drive doesn't show up with the "eject drive" option...

Works fine.

Hot swap technology is part of SATA spec, by the way. eSata cards have a
registry flag to enable the "disconnect this device" interface. Internal
SATA controllers do not. That's the only difference between the two. You
can use an eSata drive with an internal controller if you have the
appropriate SATA to eSata cable. I work with lots of SATA drives due to
my data recovery business.
